目的探讨低浓度亚砷酸钠诱导肝癌干细胞(LCSCs)分化的分子途径。方法采用免疫荧光、Western blotting和荧光定量PCR分析肝细胞癌(HCC)组织和细胞PML、Oct4和Sox2基因的表达情况,比较低浓度亚砷酸钠处理后HCC细胞PML、Oct4和Sox2基因表达及LCSCs生物功能的变化,确定低浓度亚砷酸钠对LCSCs分化的诱导作用及其分子途径。结果 0.5μg/ml亚砷酸钠可明显抑制Hu H7和原代HCC细胞形成肿瘤球,增强Hu H7细胞对吡柔吡星(THP)的敏感性(P〈0.01),并抑制Hu H7和原代HCC细胞移植成瘤的能力(P〈0.05)。HCC组织和细胞表达PML、Oct4、Sox2m RNA和蛋白,且0.5μg/ml亚砷酸钠可使Oct4、Sox2 m RNA(P〈0....
