PTEN defects underlie disorders with several heritable neoplasms, including the Cowden syndrome, but many patients with these syndromes do not carry
PTEN
mutations. Some patients with hamartomatous inherited tumors have enhanced function of the WWP1 ubiquitin ligase that increases PTEN degradation and fosters tumor-promoting PI3K activity.
