R587.2; 目的 探讨小鼠高糖记忆模型中足细胞向上皮间质细胞转化(EMT)的机制.方法 将小鼠肾小球足细胞分为4组:正糖组(5.5 mmol/L D-葡萄糖)、高糖组(30 mmol/L D-葡萄糖)、记忆组(先30 mmol/L D-葡萄糖,后5.5 mmol/L D-葡萄糖)、渗透压组(5.5 mmol/L D-葡萄糖+24.5 mmol/L甘露醇),除记忆组作用96 h外,其余组均作用48 h.Western blotting及免疫荧光检测各组足细胞EMT指标肾病蛋白(Nephrin)及α-平滑肌肌动蛋白(α-SMA)的表达.采用Illumina HiSeq 2000进行正糖组、高糖组及记忆组的转录组测序,DESeq鉴定差异表达基因,并对差异表达基因进行GO及KEGG富集分析.最后...
