With reconstituted in vitro systems and live cell experiments using optogenetic motor recruitment, we asked how KIF21B motors move with a retrograde bias on mixed polarity microtubule networks. Plus end–out MTs were more dynamic than plus end–in MTs in dendrites. These native MT dynamics contribute to the retrograde bias of cargo movement.
